Determination of multi-bioactive compounds in the root of Ricinus communis using HPLC- Evaporative Light Scattering Detection (HPLC-ELSD) method for quality control.

Abstract

First time, a simple, and reliable method based on HPLC coupled to an evaporative light scattering detector (ELSD) was established for the simultaneous determination of triterpenes and phytosterols viz. erandone (urs-6-ene-3,16-dione) (1), lupeol (2), stigmasterol (3) and sitosterol (4) in Ricinus communis (Erand) root, an important traditional medicinal plant used in Indian System of medicine. The chromatographic separation was performed on a Phenonemex Luna-C18 (4.6mm × 50mm, 3?m) using isocratic elution of 1% acetic acid in acetonitrile and 1% acetic acid in water. Diverse method validation parameters, such as the linearity (r > 0.999), sensitivities (LOD and LOQ), accuracy, precision, repeatability, and reproducibility, were successfully obtained. These four phytochemicals (1-4) including one new diketone pentacyclic triterpene (erandone) show potential as marker compounds for evaluating the quality of R. communis and the variation in its chemical composition due to geographical-environmental interaction.
